Internally Funded PhD Student (m/f/d) - Cellular senescence in cancer and bone metastasis (Saul group), (65%)Full-timeTätigkeitsfeld: Wissenschaft und ForschungBeschäftigungsumfang: VollzeitCompany DescriptionThe research group of Prof. Dominik Saul is part of the Robert Bosch Center for Tumor Diseases (RBCT). The RBCT is a newly founded non-profit translational cancer research institute at the Bosch Health Campus (BHC) in Stuttgart, in direct proximity to the Robert Bosch Hospital (RBK). As a part of the National Center for Tumor Diseases (NCT) Southwest and the Comprehensive Cancer Center Tübingen-Stuttgart, the RBCT is dedicated to innovative cancer research and the development of new treatment strategies. The Saul group has a particular interest and expertise in cellular senescence. In particular, we utilize state-of-the-art techniques such as various genome-/transcriptome-wide sequencing approaches, multiplex immunofluorescence staining of tissues as well as genetic engineering and other work on tumor cell lines. Our main research goal is to explore cellular senescence and the senescence-associated secretory phenotype (SASP) in cancer cells and the tumor microenvironment during tumor development, progression and metastasis. We aim to decipher underlying molecular mechanisms in order to identify novel therapeutic targets to improve treatment response.Group description: : https://www.rbct.de/en/cellular-senescence-cancer-and-bone-metastasisJob DescriptionUtilize modern sequencing-based molecular biology approaches (RNA-seq, scRNA-seq, ATAC-seq, ChIP-seq) to examine transcriptomic and gene regulatory mechanisms controlling tumor- and senescent cell identity and therapy response.Use bioinformatic analyses and interpretation of data to perform hypothesis testing and generation.Perform multiplex immunofluorescence staining to confirm gene regulatory mechanisms in primary tumor tissue using multispectral imaging with an Akoya PhenoImager, perform Telomere-associated DDR foci (TAF) staining.Perform tissue-based preparation of histological samples (FFPE tumor material) for single cell RNA-sequencing using the 10X Genomics technology.Perform advanced cell culture experiments to examine the effects of drug treatment in various translational model systems.Participation in internal or external research collaborations.Documentation of work procedures, data acquisition and analyses.QualificationsApplicants must have completed a Master of Science in a related field (biology, molecular biology, biotechnology, bioinformatics or similar)Experience with cell culture, transfections, stainings, etc.Extensive molecular biology experience (real-time PCR, Western blot, etc.)Experience with bioinformatic analyses (R, Python)Strong work ethic and high level of self-motivationIndependent, goal-oriented and structured way of working and ability to work in a teamConfident written and spoken English skillsAdditional InformationWe offer you:An exciting and international research environmentExcellent experience and expertise in modern epigenetic methodsOutstanding bioinformatic expertiseExtensive modern infrastructure for all methods related to the projectWorking in a committed team, characterized by mutual appreciation and supportLong track record in bone research and bioinformatics (see https://pubmed.ncbi.nlm.nih.gov/?term=Saul+Dominik&sort=date :This position is not contingent upon the applicant having attained or attaining an externally funded fellowship for the duration of their project.Contact:Prof. Dr. Dominik Saul Group Leader E-Mail: [email protected] Vita: https://www.rbct.de/en/team/dr-dominik-saulSind Sie bereit für eine neue Herausforderung?
